Diabetes Mellitus (DM) is a metabolic disease that developed due to the pancreas does not sufficient produce insulin or body cannot effectively use it produces. Genetic factors have an essential role in development of Type 2 (DMT2), which impaired production by pancreatic β cells, resistance, and action. The single nucleotide polymorphisms (SNP) KCNJ11 rs5219 affected cell activity can inhibit release, thus causing decrease therapeutic effectiveness. purpose this study identify SNP gene measure patient blood sugar levels as outcome therapy. A cross-sectional was conducted prospectively at Moewardi Hospital, Surakarta, involving 10 patients with DMT2 who received sulfonylureas DNA isolated from whole sample patients. PCR amplification performed amplify gene, followed sequencing. 2-H PP, FPG , HbA1c parameters were measured outcomes. results showed genotype frequencies (AA-AG-GG) 10%, 50% 40%, while allele frequency (A-G) 35% 65%. uncontrolled values for 2H-PP on (AA AG + GG) 10% 20%; genotypes AA GG 40%; 80%.
